Immunohistochemistry of formalin-fixed, paraffin-embedded human colon carcinoma using S0391-10, which was peroxidase-conjugated to the secondary antibody, followed by DAB staining. This data demonstrates the use of S0391-10 for immunohistochemistry.

Fluorescent confocal image of HeLa cells using S0391-10. HeLa cells were fixed with 4% PFA (20min), permeabilized with Triton X-100 (0.2%, 30min). Cells were then incubated with S0391-10 (1:100, 2hr at RT). For secondary antibody, Alexa Fluor® 488 conjugated donkey anti-rabbit antibody (green) was used (1:1000, 1hr). Nuclei were counterstained with Hoechst 33342 (blue) (10ug/ml, 5min).

Overlay histogram showing K562 cells stained with S0391-10 (green line). The cells were fixed with 2% paraformaldehyde (10min) and then permeabilized with 90% methanol for 10min. The cells were then icubated in 2% BSA to block non-specific protein-protein interactions followed by S0391-10 (1:25) for 60 min at 37ºC. The secondary antibody used was Goat-Anti-Rabbit IgG, DyLight® 488 Conjugated Highly Cross-Adsorbed (1:200) dilution for 40min at 37ºC. Isotype control antibody (blue line) was rabbit IgG1 (1ug/1x10e6 cells) used under the same conditions. Acquisition of >10,000 events was performed.
